چکیده

Wiley, W. R. (Washington State University, Pullman) and J. L. Stokes. Requirement of an alkaline pH and ammonia for substrate oxidation by Bacillus pasteurii. J. Bacteriol. 84:730-734. 1962-Resting-cell suspensions of Bacillus paseurii require a pH higher than 8.0 and also NH(4) (+) for the oxidation of glycine, alanine, serine, glutamic acid, other amino acids, and compounds of the Krebs cycle. The optimal pH is 9.2. Maximal activity was obtained with 0.5% (NH(4))(2)SO(4), the highest concentration tested. NH(4) (+) can be supplied by a number of inorganic and organic ammonium salts. The effect of NH(4) (+) is fairly specific. It cannot be replaced by Na(+), Cs(+), and Mg(++), and only partially replaced by Li(+), K(+), Rb(+), and Ca(++). Some possible metabolic functions of the required alkaline pH and NH(4) (+) are discussed.

Effect of ammonia on the growth of Bacillus species and some other bacteria.

The tolerance of 26 Bacillus species isolated from alkaline fermented foods, five other bacilli and nine non spore-forming bacteria to alkaline pH and ammonia was determined. All grew at pH 7, 8 and 9 in the presence of 930 mmol l-1 NH4 + at pH 7.0, and in the presence of NH3 concentrations up to 5 mmol l-1 at pH 7.0 and 8.0. At higher NH3 concentrations, growth of some of the bacteria was inhi...

Response of surface optimization for the enhanced production of alkaline protease isolated from Bacillus sp. with bean husk as a new substrate

Optimization of the fermentation medium for maximum alkaline protease production was carried out. Fifteen positive isolates were examined for their extent of alkaline protease production. The most potent producer was identified as Bacillus sp. The solid substrate screening showed that the combination of wheat straw and bean husk was the best one. The initial screening by using Plackett–Burman’s...

Batch Kinetics and Modeling of Alkaline Protease Production by Isolated Bacillus sp. (RESEARCH NOTE)

The aim of this study was the use of fish waste hydrolysate (FWH) as a substrate for alkaline protease production using isolated Bacillus sp. in a batch system. Then the fermentation kinetics of enzyme production was studied. The results show that with the addition of FWH to the fermentation medium with a final concentration of 4% (optimal concentration), alkaline protease value reached a maxim...

Enhancing Production of Alkaline Polygalacturonate Lyase from Bacillus subtilis by Fed-Batch Fermentation

Alkaline polygalacturonate lyase (PGL, EC 4.2.2.2) is an enzyme used in many industries. We developed a fed-batch fermentation process that combines the enzymatic pretreatment of the carbon source with controlling the pH of the fermentative broth to enhance the PGL production from Bacillus subtilis 7-3-3 to decrease the production cost. Maintaining the fermentation broth at pH 6.5 prior to feed...

Dark Hydrogen Fermentation From Paper Mill Effluent (PME): The influence of Substrate Concentration and Hydrolysis

Paper mill effluent (PME) was used as an organic feedstock for production of biohydrogen via dark fermentation using heat-shock pretreated anaerobic sludge under mesophilic conditions. The influence of substrate concentration (5, 10 and 15 g-COD/L) and the initial pH (5 and 7) on the efficiency of dark hydrogen fermentation from PME were investigated. The highest hydrogen yield of 55.4 mL/g-COD...
